Abstract: Cancer arises in the context of an in vivo tumor microenvironment. This microenvironment is both a cause and consequence of tumorigenesis. Tumor and host cells co-evolve dynamically through indirect and direct cellular interactions, eliciting multiscale effects on many biological programs, including cellular proliferation, growth, and metabolism, as well as angiogenesis and hypoxia and innate and adaptive immunity. Here we highlight specific biological processes that could be exploited as targets for the prevention and therapy of cancer. Specifically, we describe how inhibition of targets such as cholesterol synthesis and metabolites, reactive oxygen species and hypoxia, macrophage activation and conversion, indoleamine 2,3-dioxygenase regulation of dendritic cells, vascular endothelial growth factor regulation of angiogenesis, fibrosis inhibition, endoglin, and Janus kinase signaling emerge as examples of important potential nexuses in the regulation of tumorigenesis and the tumor microenvironment that can be targeted. We have also identified therapeutic agents as approaches, in particular natural products such as berberine, resveratrol, onionin A, epigallocatechin gallate, genistein, curcumin, naringenin, desoxyrhapontigenin, piperine, and zerumbone, that may warrant further investigation to target the tumor microenvironment for the treatment and/or prevention of cancer.

Abstract: Opuntia flowers are a natural source of biologically active compounds and they have been used as medicinal plant for a long time. Despite the various uses reported for the decoction and infusion of these flowers, their characterization has been discarded. In this study, the decoction and infusion prepared from Opuntia ficus-indica were analyzed with respect to their content in minerals and phytochemicals in order to evaluate its nutritional characteristics. The obtained data proved that these preparations are a rich source of minerals mainly K and Ca. Moreover, the phytochemical analysis revealed that they have important polyphenols, flavonoids and tannins contents with the infusion that presented the highest polyphenol levels. LC-MS analyses of decoction and infusion allowed the characterization of 20 phenolic compounds. It is mainly identified by the presence of flavonols glycosides.

Abstract: Our previous studies demonstrated that the combination of treatment with ascorbic acid (AsA) and X‑ray irradiation results in increased apoptosis in HL60 cells. The present study was performed to investigate the effects of the combined use of AsA and X‑ray irradiation on epithelial cancer and sarcoma cells, and its potential use in future clinical treatment. X‑ray irradiation combined withAsA treatment resulted in increased suppression of cell growth of HT1080, SAS and A549 cells in vitro compared with X‑ray irradiation alone. The combined treatment also suppressed tumor growth in implanted HT‑1080 cells in vivo. Using annexin V/propidium iodide staining and the detection ofactivated caspase 3, it was found that X‑ray irradiation increased the apoptotic rate of HT1080 cells and resulted in G2/M arrest. However, apoptosis in the HT1080 cells treated with 5 mM AsA remained unchanged, and no changes were observed in the G2/M fraction. By contrast, AsA treatment caused increased suppression of proliferation compared with X‑ray irradiation. These results suggested that 5 mM AsA slowed the cell cycle and reduced tumor growth. Therefore, X‑ray irradiation combined with AsA treatment may be effective against epithelial cancer and sarcoma cells.

Polyphenols Target Fusobacterium nucleatum and the Host Inflammatory Response: Potential Innovative Molecules for Treating Periodontal Diseases. Abstract: Blueberries contain significant amounts of flavonoids to which a number of beneficial health effects in humans have been associated. The present study investigated the effect of a polyphenol-rich lowbush blueberry (Vaccinium angustifolium Ait.) extract on the two main etiologic components of periodontitis, a multifactorial disorder affecting the supporting structures of the teeth. Phenolic acids, flavonoids (flavonols, anthocyanins, flavan-3-ols), and procyanidins made up 16.6, 12.9, and 2.7% of the blueberry extract, respectively. The blueberry extract showed antibacterial activity (MIC = 1 mg/mL) against the periodontopathogenic bacterium Fusobacterium nucleatum. This property may result from the ability of blueberry polyphenols to chelate iron. Moreover, the blueberry extract at 62.5μg/mL inhibited F. nucleatum biofilm formation by 87.5 ± 2.3%. Subsequently, the ability of the blueberry extract to inhibit the NF-κB signaling pathway in U937-3xκB cells was investigated. The blueberry extract dose-dependently inhibited the activation of NF-κB induced by F. nucleatum. In addition, a pretreatment of macrophages with the blueberry extract (62.5 μg/mL) inhibited the secretion of IL-1β, TNF-α, and IL-6 by 87.3 ± 1.3, 80.7 ± 5.6, and 28.2 ± 9.3%, respectively, following a stimulation with F. nucleatum. Similarly, the secretion of MMP-8 and MMP-9 was also dose-dependently inhibited. This dual antibacterial and anti-inflammatory action of lowbush blueberry polyphenols suggests that they may be promising candidates for novel therapeutic agents.

Broccoli Extract Does The “Impossible”: Improves Autism Spectrum Disorder
Of all the natural substances we have reviewed with potential value in the treatment of ASD, sulforaphane, a molecule found in cruciferous vegetables such as broccoli, cabbage and Brussels sprouts, is perhaps the most promising candidate.
A groundbreaking study published in 2014 in The Proceedings of the National Academy of Sciences USA titled, “Sulforaphane treatment of autism spectrum disorder (ASD),” found that a broccoli sprout extract significantly improved the behavior of boys and men with ASD. The researchers pointed out that, presently, no conventional, mechanism-based treatment is available for ASD. Sulforaphane was selected, in part, because its physiological effects are well characterized and ideal for those with ASD:
Dietary sulforaphane, of recognized low toxicity, was selected for its capacity to reverse abnormalities that have been associated with ASD, including oxidative stress and lower antioxidant capacity, depressed glutathione synthesis, reduced mitochondrial function and oxidative phosphorylation, increased lipid peroxidation, and neuroinflammmation.”
The placebo-controlled, randomized pilot study of 44 males, ages 13-27, showed that after 18 weeks of treatment with a sulforaphane-rich broccoli sprout extract, 46% had significant improvements in social interactions and 42% has significant improvements in verbal communication.
Additionally, more than half the participants showed decreases in abnormal behaviors, including irritability, hyperactivity and repetitive movements. Of note, once treatment stopped, most of the behaviors returned to pretreatment levels. The dosing schedule was determined by body weight:
- 100 lbs or less: one capsule containing 50 µmol (232 mg) of sulforaphane-rich broccoli extract was given daily
- 101–199 lbs, 100 µmol (two capsules of 232 mg each) of sulforaphane-rich broccoli extract was given daily
- More than 200 lbs: 150 µmol (three capsules of 232 mg each) of sulforaphane-rich broccoli extract was given daily
